Abstract : This research proposal aims to prepare biosensing surfaces through modification of crystalline silicon surfaces by covalent attachment of organic monolayers. These nanoscaled organic films allow reversible and selective adsorption of biomolecules such as proteins, enzymes and antibodies, which are very useful parts in biosensing devices. Mixed organic monolayers of functionalized and non-functionalized alkenes will be formed on cleaned oxide free hydride-terminated crystalline silicon surface. The hydride-terminated crystalline silicon surface will be formed by the reaction of dilute solutions of hydrofluoric acid on chemically cleaned silicon surfaces. Organic monolayers will be formed by boiling the H-terminated silicon surface with neat functionalized and non-functionalized alkenes or their solutions in a give aromatic solvent under inert atmosphere. Hydrolysis of the mixed monolayer allows the formation of specific functional groups which can be reacted with amino compounds. The reaction affords an amide-terminated nanospots. Based on the length of the alkyl chain attached to the silicon surface, specific adsorption of certain biomolecules will take place selectively and hopefully reversibly. The formed monolayers will be characterized by the static water contact angle, infrared spectroscopy and x-ray photoelectron spectroscopy. Adsorption of proteins will be measured by atomic force microscopy.
